In the intrinsic pathway, caspase activation requires the formation of an activating complex called an apoptosome. This complex is made up of four components, including cytochrome c, apoptosis-activating factor 1 (Apaf-1), adenosine triphosphate (ATP), and procaspase-9.
What are Procaspases?
Procaspases are the inactive precursors to caspases. They are activated by cleaving one or two specific aspartic acids. The procaspase is split into two subunits that form a dimer. Two of these dimers combine to form the activated tetramer that is a caspase. This process is activated by already active caspases.
How is an apoptosome created?
The apoptosome is a large quaternary protein structure formed in the process of apoptosis. Its formation is triggered by the release of cytochrome c from the mitochondria in response to an internal (intrinsic) or external (extrinsic) cell death stimulus.
How are Procaspases activated?
Following release of cytochrome c from the mitochondria, procaspase-9 is recruited by its CARD to the apoptosome, causing its activation [19]. This is hypothesized to occur by bringing inactive caspase-9 monomers into a high local concentration, allowing dimerization to occur, which induces their activation [11].
