This is caused by killing Ysolda before beginning the drinking contest, since without her, you cannot find out where Sam went. However, you will not receive a “Quest Failed” notification upon killing her, meaning the quest cannot be started. Marrying Ysolda may fix this. (Resolved With Skyrim Patch 1.5)
