What is Petrifilm method?

Petrifilm plates are designed to be as accurate as conventional plating methods. Ingredients usually vary from plate to plate depending on what micro-organism is being cultured, but generally a Petrifilm comprises a cold-water-soluble gelling agent, nutrients, and indicators for activity and enumeration.

How do Petrifilm plates work?

Petrifilm™ plates are designed to determine total aerobic bacterial populations. The easy-to-use Petrifilm plates contain a film coated with nutrients and gelling agents. You will be able to monitor environmental bacterial counts using a variety of methods.

How do I reduce my aerobic plate count?

Exposure of eggshells to 30 and 60 s UV significantly reduced aerobic plate counts compared to untreated eggs. Exposure to 60 s of UV resulted in a 2 to 3 log10 cfu/egg APC reduction and reduced counts below detectable levels.

What is the correct countable range for a Petrifilm count?

1-300
APC 3M Petrifilm plates are incubated at 35 ± 1C for 48 ± 3 h. Count all colonies in the countable range (1-300).

What are two advantages of Petrifilm plates over petri dishes?

The advantages

  • 85% less space than the classic agar.
  • 45% less work.
  • 80% increased efficiency.
  • 1/2 faster data.

How do you dispose of 3M Petrifilm?

As a disposal alternative, incinerate in a permitted waste incineration facility. If no other disposal options are available, waste product may be placed in a landfill properly designed for industrial waste. For Transport Information, please visit http://3M.com/Transportinfo or call 1-800-364-3577 or 651-737-6501.

What does E coli look like on Petrifilm?

Confirmed coliforms are red or blue colonies with associated gas bubbles. Confirmed E. coli are blue colonies with associated gas bubbles. Results are fast and accurate.

What is a good aerobic plate count?

Counts outside the normal 25-250 range may give erroneous indications of the actual bacterial composition of the sample. Dilution factors may exaggerate low counts (less than 25), and crowded plates (greater than 250) may be difficult to count or may inhibit the growth of some bacteria, resulting in a low count.

What causes high aerobic plate count?

Large numbers of bacteria may be an indication may be an indication of poor sanitation or problems with process control or ingredients. Certain products, such as those produced through fermentation, naturally have a high APC.

What is coliform Petrifilm?

The 3M Petrifilm Coliform Count Plate is a sample-ready-culture-medium system which contains Violet Red Bile (VRB) nutrients, a cold-water-soluble gelling agent, and a tetrazolium indicator that facilitates colony enumeration in food and beverage samples.

What is APC Petrifilm?

Aerobic Plate Count (APC)1 3M Petrifilm contains nutrients and 2,3,5-triphenyltetrazolium chloride as an indicator of bacterial growth. Reduction of triphenyltetrazolium by bacteria results in red coloured colonies2. Plates are hydrated with sample and gelling agents cause the media to solidify.

What is the advantage of using 3M Petrifilm as compared to the traditional agar petri dishes?

Unlike traditional agar methods, the 3M™ Petrifilm™ Plates are ready to use – no preparation required.

What is the recommended sample pH for 3M Petrifilm lactic acid bacteria count plate?

> 5
Considering the low pH of these products, you may find using the 3M Petrifilm Rapid E. coli/Coliform Count Plate method a better fit for this application as the recommended pH range is > 5 and the amount of sample pH adjustment that would need to be done is minimal compared to the standard plate.

How do you store Petrifilm?

Store unopened 3M Petrifilm RAC Plate pouches refrigerated or frozen (-20 to 8°C / -4 to 46°F). Just prior to use, allow unopened pouches to come to room temperature before opening (20-25°C / <60% RH).

How do you count 3M Petrifilm?

3M Petrifilm Aerobic Count Plate with a few bacterial colonies. When colonies number more than 300, estimate the count. Determine the average number of colonies in one square (1cm2) and multiply it by 20 to obtain the total count per plate.

How do you count coliform Petrifilm?

Estimated total coliform count = 220 Estimates can be made on plates containing greater than 150 colonies by counting the number of colonies in one or more representative squares and determining the average number per square. Multiply the average number by 20 to determine the estimated count per plate.

What is the difference between total plate count and aerobic plate count?

The APC term stands for aerobic plate count, but again is interchangeable with the others. Other terms used more historically are Standard Plate Count, Mesophilic Count or Total Plate Count these too generally refer to aerobic bacteria able to grow at average temperatures (e.g. 30 to 40°C).

What is acceptable CFU?

Products intended for consumption in their raw form should contain less than 100 CFU/gram.

What is the 3M™ Petrifilm™ spreader?

This 3M™ Petrifilm™ Spreader is an easy-to-use tool designed to quickly and evenly distribute inoculum within the 3M™ Petrifilm™ Plate.

Why 3M Petrifilm™ rapid yeast and mold count plate?

3M™ Petrifilm™ Rapid Yeast and Mold Count Plate features a new indicator technology that makes colonies easier to interpret. This sample-ready-culture-medium system provides results in as little as 48 hours of incubation time.

What is 3M Petrifilm aerobic count plate?

The 3M™ Petrifilm™ Aerobic Count Plate is a ready-made culture medium system that contains modified Standard Methods nutrients, a cold-water-soluble gelling agent and an indicator that facilitates colony enumeration. 3M Petrifilm Aerobic Count Plates are used for the enumeration of aerobic bacteria.
